Unique Identifier : AIDSLINE ASM94/94313115 AB Pneumocystis carinii contains a large (90-120 kDa) surface glycoprotein that is recognized by antibodies in the serum of immune and infected hosts and seems to play a role in binding to host cells. Recent genetic analysis of MSG cDNAs has shown that MSG is likely to be a family of proteins. A fragment from the genome of rat-derived P. carinii was found to contain two MSG genes arranged as a direct repeat. The sequences from one gene (MSG B), the region between the two genes, and part of the second gene (MSG A) were determined. The open reading frames of MSG A and MSG B encode non-identical proteins, both of which are similar to that encoded by a previously published cDNA. The 5' and 3' untranslated regions of the MSG gene pair were highly conserved. Genetic analysis of MSG genes and cDNAs has assisted in defining presumptive variable and constant regions of the MSG proteins, which can then be utilized in examining the expression of this protein family. In another approach to define variable and constant regions, an expression library of rat-derived P. carinii was screened with monoclonal antibodies that recognize MSG from rat-derived and human-derived P. carinii. Some of these monoclonal antibodies appear to recognize an epitope that varies among MSG proteins, while two others may recognize epitopes that are conserved among these proteins.
