To evaluate the comparative immunoreactivity of synthetic peptide (SP) fragments from immunodominant region of gp41 HIV-1 for the greater SP-based ELISA sensitivity. METHODS. Antigens: SP(1)aa584-604, Wang, 1986; (2)aa601-624, linear; (3)aa601-624, cyclic, with C603-C609 S-S bond; Sera panels: A (29 WB confirmed HIV-1 positive samples from patients followed-up at the Inst.Imunol. since 1985); B (16 WB confirmed HIV-1 positive samples with low antiHIV antibody titers, lycensed in RF for quality control of commercial HIV ELISAs), C (11 WB confirmed HIV-1 positive samples, Uganda). RESULTS. The ratios of # samples tested/# samples detected as positives were for panel A 23/29 (SP1), 28/29 (SP2), 29/29 (SP3); for panel B 0/16 (SP1), 9/16 (SP2), 16/16 (SP3); for panel C 4/11 (SP1), 6/11 (SP2), 10/11 (SP3). DISCUSSION AND CONCLUSIONS. Cyclic SP3 provides more accurate HIV Ab detection, than its linear analog (SP3 recognizes 55 of 56 specimens tested vs 43 of 56 for SP2), especially for the weak (low Ab titer) sera, probably due to better modelling of the At-binding site of viral gp41.
