Unique Identifier : AIDSLINE ICA10/94369531 AB OBJECTIVE: To find out good surrogate markers for analysis of pathophysiology and prediction of prognosis of HIV disease. SUBJECTS AND METHODS: Absolute counts of lymphocyte subpopulations (CD4 x CD45RA, CD4 x CD29, CD8 x CD11b, CD16 x CD56) were analyzed by two-color flowcytometry using Cytron-Absolute (Ortho) in 74 patients with HIV infection (CDC category A, B, C). In 8 patients, time courses of absolute counts of subpopulations were evaluated. Half of the patients were hemophiliacs. RESULTS: 1) CD4 counts measured by Cytron-Absolute seemed to be more reliable than those obtained through conventional method by calculation as WBC x lymphocyte (%) x CD4 (%). 2) Absolute counts of all of CD4+ subpopulations (CD4+CD45RA+, CD4+CD45RA-, CD4+CD29+, CD4+CD29-) declined almost parallel during the course of the desease. 3) Absolute counts of CD8+CD11b- (Tc) declined more slowly than CD4+ counts did. Opportunistic infections (OIs) frequently observed when CD8+CD11b- counts declined to a level below 200 cells/microliters. Thus, CD8+CD11b- counts may be a better surrogate marker for probability of OIs than CD4+ counts. 4) Absolute counts of CD16+CD56- NK cells stayed relatively constant when compared with CD16+CD56+ NK cells which declined steadily with time. CONCLUSIONS: Preliminary study on absolute counts of lymphocyte subpopulations was carried out. Some of the subpopulations seemed to be useful for the analyses of pathophysiology and prognosis of HIV disease.
