Unique Identifier : AIDSLINE ICA10/94369429 AB OBJECTIVE: Human Herpesvirus 7 (HHV-7), a new T-lymphotropic herpesvirus, is related to, but significantly different from HHV-6. HIV-1 and HHV-7, as well as HHV-6, are believed to share CD4+ T lymphocytes as major target cells, and, for this reason, we studied the interaction of HIV-1 and HHV-7 using a CD4+ T cell line. METHODS: A CD4+ CD8+ cell line, SUP-T1, and a HIV carrier cell line established from SUP-T1 were tested for permissiveness to HHV-6 or HHV-7 replication. RESULTS: The SUP-T1 is to date the only cell line, among 10 CD4+ T cell lines, susceptible to the productive infection of HHV-7. Expression of the CD4 antigen was completely downregulated in the HIV-1 carrier cell line. While superinfection of HIV-1 carrier SUP-T1 cells with HHV-6 was readily accomplished, the same cells were completely resistant to superinfection with HHV7. Furthermore, treatment of SUP-T1 cells with anti-CD4 monoclonal antibodies inhibited the growth of HHV-7. On the other hand, HHV-7 carrier SUP-T1 cells were readily superinfected with HIV-1. DISCUSSION AND CONCLUSIONS: Resistance of HIV-1 carrier SUP-T1 cells to HHV-7 is due to the absence of the CD4 antigen on the cell surface. HIV-1 and HHV-7 are supposed to use the same molecule, the CD4 antigen, as the virus receptor.
