Unique Identifier : AIDSLINE ICA10/94369379 AB OBJECTIVE: The purpose of the investigation was to analyze the dynamic interaction between disease progression and lymphocyte subsets in the blood and lymph nodes (LN). METHODS: The time courses of changes in CD4 subsets and CD4/CD8 ratios were compared in bioptic samples of the LN and blood from SIVmac239 infected rhesus and cynomolgus monkeys. RESULTS: Although an apparent decrease of CD4 cells, especially CD29high CD4 cells, was recognized in the blood. the percentages of CD4 cells and CD4/CD8 ratios in the LN did not decline but rather increased at several times after SIV infection. In the LN, the CD29lowCD45RAlow CD4 subset appeared after virus inoculation and became predominant as the lesions of the LN progressed. This uncommon CD4 subset was inhibited from circulating to the periphery, resulting in a deficiency of the CD29high and CD45RAhigh CD4 cell pool in the LN. DISCUSSION AND CONCLUSIONS: These qualitative changes of the CD4 cells in the LN might be one of the reasons for the decline in circulating CD4 cells.
