Unique Identifier : AIDSLINE ICA10/94369366 AB OBJECTIVE: To elucidate the mechanism of edema in AIDS-associated Kaposi's sarcoma (AIDS-KS) lesions. METHODS: Long-term cultured AIDS-KS spindle cells and their semi-purified conditioned medium (AIDS-KS CM) through heparin sepharose affinity column were tested to detect the activity inducing vascular hyper-permeability (VHP) in animal skin and promoting vascular endothelial cell growth in vitro. To identify the active agent produced by AIDS-KS cells, Western blot was done by using each fraction from the affinity column with anti-vascular endothelial growth factor(VEGF165) antibody and Northern blot was done by using RNA extracted from various AIDS-KS cell isolates with VEGF cDNA probe respectively. RESULTS: Both cultured AIDS-KS cells and AIDS-KS CM induced a specific histamine independent VHP in animal model and AIDS-KS CM demonstrated endothelial cell growth promoting activity in vitro. Two bands of 23.5 kD and 21.5 kD were identified in 0.5 M NaCl fraction by Western blot and high levels of VEGF mRNA in all AIDS-KS cell isolates by Northern blot. Moreover, the biological activity was totally neutralized by antibody to VEGF165. CONCLUSION: VEGF or VEGF like molecule was identified as the active agent causing significant complication of the disease: edema in AIDS-KS lesions.
