Unique Identifier : AIDSLINE ICA10/94369364 AB OBJECTIVE: To characterize on the molecular level a new class of HIV-associated lymphomas where the retrovirus HIV has inserted non-randomly into the tumor genomic DNA material. METHODS: 1) frozen section and paraffin section immunohistochemistry; 2) Southern blot analysis of inverse PCR products; 3) direct PCR sequencing; 4) mRNA analysis (Northern blots, cDNA-PCR). RESULTS: Inverse PCR (iPCR) uses the ability of restriction digests (Sau3a), circularization, and long terminal repeat primers to capture flanking sequences of the inserted retrovirus. Direct sequencing showed insertion of retroviral LTR sequence in exon Z of the fur fps/fes gene complex. The PCR products were subsequently probed with LTR and fur oligo's on Southern blots. TABULAR DATA, SEE ABSTRACT VOLUME. CONCLUSIONS: Fur fps/fes is a proto-oncogene complex (tyrosine kinase) activated in some myeloid tumors. The non-random (4 cases) clonal integration of HIV near this locus and subsequent differential activation of mRNA compared to control tissue (1 case) indicates that these lymphomas may have undergone retroviral insertional mutagenesis.
