Unique Identifier : AIDSLINE ICA10/94369282 AB OBJECTIVE: The purpose of the studies is to clearly determine the protein myristoylation of the gag protein in HIV-1 infected cell lines, and to resolve the role of the gag protein myristoylation in virion particle formation. METHODS: HIV-1 producing cell lines (CEM/LAV-1, U937/IIIB, U937/RF, H9/MN, Molt-4/IIIB, U1, ACH-2) were maintained at 37 degrees C in RPMI-1640 medium (10% FCS) in 5% CO2. The N-myristoylation of the gag protein was determined both by fluorography with [3H]myristic acid and by western immunoblotting analyses with anti N-myristoyl-Gly monoclonal antibody. The role of the N-myristoylation was resolved by fluorography, immunochemical and paper technological methods with N-myristoyl inhibitors or cross-linkers. RESULTS AND DISCUSSION: The N-myristoylation of the gag protein in HIV-1 producing cell lines was reconfirmed and its blockage by the myristoyl inhibitors resulted in depression of the product of HIV-1 progeny virus. The cross-linked proteins (p17gag-gp41 and p17gag-gp160) were clearly detected by western immunoblotting analyses after cocultivation of CEM/LAV-1 in the presence of cross-linking reagents. These results suggest that the N-myristoyl moiety of the gag protein may be important for maturation of infectious viruses.
