Unique Identifier : AIDSLINE ICA10/94369256 AB Production of viral antigens and infectious virus by Jurkat cells transfected with pHXB2 (nef+) progressively declined over an extended period of time in culture. Sixty clones were generated from the transfected line; they were analyzed for the presence of provirus and monitored for the expression of viral antigens (surface gp120 and p24) and CD4. Six/60 clones were positive for the release of p24. Circular viral DNA was present in all p24 positive clones and absent in all p24 negative clones (54/60). In addition to circular DNA, 5/6 p24+ clones contain an altered form of the original construct. The remaining p24+ clone had the original pHXB2 gpt construct, as well as an altered form of the construct; it releases high levels of p24. All the p24- clones (54/60) contained altered forms of pHXB2 (but not the original construct). No clonally integrated provirus could be detected in any of the clones, suggesting that the clonal progenitor cell did not contain an integrated provirus. Thus, in the absence of integration, virus may persist in an unintegrated form, yet still be expressed and result in an active infection.
