Attenuation was effected by the precise deletion of 18 open reading frames associated with virulence or host range. NYVAC is safe by numerous tests including intracranial inoculation into newborn mice. No disseminated infection was observed in severely immunocompromised animals. ALVAC is an avipoxvirus, canary-pox. Avipoxviruses are restricted for productive viral replication to avian species and in mammals provide uniquely safe immunizing vehicles. Upon inoculation in mammalian species, including man, ALVAC elicits cellular, humoral and protective immunity. One of the features inherent within this vector approach is the intracellular synthesis of antigens which allows for the association of peptides with MHC-1. Cellular immune responses including specific induction of CD8+ CTL responses have been demonstrated with human immunodeficiency virus, cytomegalovirus and rabiesvirus proteins in laboratory animal model systems as well as in man. These vectors are also useful in the ex vivo expansion of CD8+ CTLs from HIV seropositives. The utility of these vectors for the expression of tumor-associated antigens in conjunction with specific cytokines is just now beginning to be evaluated.
