Unique Identifier : AIDSLINE ASHM5/94348921 AB Transcriptional regulation involves the interaction of enhancer-promoter regions with cellular binding proteins. NF-kB is a nuclear transcriptional factor which binds and mediates transcription from the sequences in the promoter region. In our system we have studied NF-kB and related proteins in PHA stimulated cord blood mononuclear cells. In this model the cells were treated with rIL2 and with or without DEAE-dextran, and nuclear extracts were made. Binding of NF-kB enhancer region sequences with these extracts were studied by gel shift assays. As well as NF-kB, another protein NF-TIM was detected in large amounts. This NF-TIM also binds to NF-kB region even when it is mutated (GGG to CTC) but binding is decreased with double mutated enhancer (GGG to CTC and CCG to AAC). Further studies of the role of NF-TIM in HIV-1 replication are in progress with UV crosslinking, footprinting, and cloning of enhancer region and its mutants in a reporter gene system.
