CD8+ T cells predominate, and epitopes exist in most viral antigens. Generation of cytotoxic T cells from the PB mononuclear cells from uninfected donors has received less attention. Our aim is to characterise the primary cellular immune response to HIV-1 infection in vitro. The approach is to generate T-cell colonies and amplify T-cell lines and clones. These T-cells are phenotyped (cell markers) and their HIV-1 specificity determined by use of autologous targets infected with HIV-1 or with vaccinia constructs expressing HIV-1 antigens. So far, 92 T-cell lines and 20 clones have been established in response to stimulation with live or inactivated HIV-1. Both CD4+ and CD8+ T cells have been generated. Cytotoxicity tests have so far yielded 6 T-cell lines with putative HIV-1 specificity for detailed study.
