Unique Identifier : AIDSLINE ICA10/94371511 AB OBJECTIVE: In the hope of exploring the possible applications of Kampo medicine for HIV infection, we examined HIV infected subjects if they had specific conformation or not. Based on our previous findings that one of a TCM, Sho-Saiko-To(SST), inhibited the production of factors which facilitate the replication of HIV, such as superoxide(SOx) and prostaglandin E2, the method to detect more sensitively and simply SOx production was further investigated. METHODS: (1) 40 HIV infected homosexual subjects (Asymptomatic 30, ARC 4, AIDS 6) were examined to diagnose the conformation for application of Kampo medicine according to the TCM diagnostic method. (2) SOx production from neutrophils stimulated with phorbol myristate acetate (PMA) and opsonized zymosan(OpZ) were determined by Flow Cytometry method using 100 microliters of whole blood. Effect of TCMs on SOx production was investigated. RESULTS AND DISCUSSION: (1) 65% of the HIV infected subjects showed the conformation of SST or combined prescription with SST. Since TCM is clinically effective when the patient has corresponding conformation, SST may be widely applicable for HIV infection. (2) SOx scavenging effect of TCM was more sensitively detected when stimulated with OpZ than with PMA, indicating this Micro-assay by Flow Cytometry may be a useful method to evaluate the effect of TCM on SOx production.
