Unique Identifier : AIDSLINE ICA10/94371414 AB OBJECTIVE: HIV is known for sexually transmited virus, but no effective method of HIV free sperm preparation from HIV carrier's semen, use for artificial insemination of husband (AIH). METHODS: On informed consent, collected HIV carrier's semens were treated 4 layer uncontinuous Percoll gradients, and each fruction of the gradients was tested for sperm count, contaminated cell count. HIV p24 antigen detection by co-cultivation with normal man's PBL. HIV provirus by PCR and HIV RNA by RT-PCR. RESULTS: From specific gravity 1110 fruction, no HIV was detected by all above detective methods and cotaminated cells were disappeared. In same fruction, sperm was prepared enough for AIH. DISCUSSION AND CONCLUSION: This sperm preparation method from HIV carrier's semen possible diminishes HIV infectivity at AIH.
