Previously we have reported that HIV envelope protein gp160 can enhance myelopoiesis in cord blood (Blood, 80: 1463, 1992). This study investigates influence of HIV gp160 in secretion of humoral CSFs: IL-3, IL-6 and GM-CSF which are essential for myelopoiesis, in cord blood. Whole cord blood (WCB) cells and purified cord blood T cells (CB-T) were treated with gp160 for 6 hours for detecting mRNA expression by RT-PCR and 24 hours for quantitation of cytokines by ELISA. All 3 CSFs were induced in CB-T maximally with significantly lower amounts in WCB cells. Treatment with sCD4 consistently abolished IL-3 and GM-CSF production from both purified CB-T and WCB cells whereas IL-6 secretion was inhibited only in CB-T but enhanced in WCB. Our studies indicate that i) gp160 is a potent inducer of humoral CSFs in cord blood ii) that the major source of these cytokines in cord blood is T cells rather than monocytes and iii) induction of these cytokines is mediated through the interaction of gp160 with CD4 molecules but a different mechanism may be involved for IL-6 secretion from CB-non T cells.
